IN SUMMARY

After iv administration of a drug, the initial plasma concentration is dependent on the administered dose and the volume of distribution

Higher doses produce higher initial drug concentrations when Vd is constant. Also, smaller Vd will result in higher initial drug concentration when the dose is constant.

The rate of decline of the plasma drug concentration is dependent on the elimination rate constant. The elimination rate constant. is dependent on the CLT and Vd, and is determined from: (CLT/Vd = k).

Larger k, means shorter half-life and faster rate of drug elimination from the body.
