Effect of half-life on the plasma drug concentration-time profile

After iv drug administration, the amount of the drug in the body and the plasma drug conc decline at a rate that determines the half life (and, the elimination rate constant )

Half life (hr)

Faster rate of drug elimination results in shorter half life

Different drugs have different half lives. The drug half life is dependent on the apparent volume of distribution and the efficiency of the eliminating organs of the drug (total body clearance).

The half life of a drug may change due to failure in the function of the organ responsible for drug elimination (e.g. kidney or liver). This can slow the rate of drug elimination resulting in longer drug half life
