Factors Affecting the Urinary
Excretion of Drugs (Cont'd)

b- Total body clearance

Examine the urinary excretion rate of drugs that have different CLT if the dose and CLR are constant.

Parameters
Dose (mg) CLΤ(L/hr) CLR(L/hr)
600 3 2
CLT (L/hr)

The rate of decline of the renal excretion rate versus time is dependent on k. Since (CLT/Vd = k), larger CLT results in faster decline in the renal excretion rate versus time plot (Vd is constant ).

Parameters
Dose (mg) CLΤ(L/hr) CLR(L/hr)
600 3 2
CLT (L/hr)

The fraction of the dose excreted in urine is determined from the ratio of CLR/CLT. After administration of similar doses, the amount of drug excreted in urine will be smaller when this ratio gets smaller.
