Effect of Vd on the plasma conc-time profile

Administration of the same dose of a drug to a group of patients who have different Vd will produce initial plasma drug conc that is inversely proportional to Vd.

If these patients have the same total body clearance, it will take longer time to clear the drug from the body of the patients with larger Vd. (i.e. larger Vd will lead to slower decline in the drug conc, and longer half life)

After administration of the same dose to patients with similar clearance, the patients with larger Vd will have lower initial plasma drug concentration, and slower rate of drug elimination (longer half life).
