Effect of dose on the plasma conc-time profile

Because the Vd of a drug is constant in a particular patient, increasing the iv dose will produce proportional increase in the initial plasma drug concentration.

Assume that different doses of a drug were administered to the same patient who has Vd = 10 L

Dose

Administration of higher doses results in higher initial plasma drug concentrations. The slopes of the lines are similar because the elimination process is dose independent (assuming first-order elimination).
