Loading Dose

The loading dose is an iv bolus dose administered at the time of starting the iv infusion to achieve faster approach to steady state.

When an iv loading dose is administered just before the start of the iv infusion, the plasma concentration will be the sum of the concentration resulting from the iv bolus and the iv infusion.

The loading dose is chosen to achieve plasma concentration similar to or very close to the steady state plasma concentration that should be achieved with the constant rate iv infusion.

Administration of a loading dose results in faster approach to steady state, however it does not affect the steady state concentration.

The steady state plasma concentration depends on the iv infusion rate and the total body clearance.
