After Completing this module you should be able to:

• Describe the physiological meaning of organ drug clearance in terms of organ blood flow, intrinsic clearance and extraction ratio.

• Describe the effect of changing the hepatic intrinsic clearance and blood flow on the efficiency of the liver for eliminating the drug (extraction ratio).

• Analyze the effect of changing the intrinsic clearance on the plasma concentration-time profile after iv and oral administration.

• Analyze the effect of changing the liver blood flow on the plasma concentration-time profile after iv and oral administration.

• Identify the characters that will make the drug elimination rate sensitive to changes in the protein binding.
