Determination of CLT

(After a single iv drug administration)

a- An iv dose of the drug is administered, then plasma samples are obtained.

Cp0

Vd =

dose

Cp0

t1/2

k =

0.693

t1/2

b- Draw the best line that goes through the samples.

c- Starting at any point on the line, find out the half life and the elimination rate constant of the drug.

d- The Y-intercept is the plasma concentration at time zero (Cp0). The volume of distribution is determined from the dose and Cp0. (Vd = Dose/Cp0).

e- The CLT is calculated from Vd and k

CLT = k Vd
