a - Plasma conc-time profile

The conc-time profile during the elimination phase is not linear on semilog scale.

The rate of drug elimination is different at
different drug conc. The half life and total
body clearance are conc-dependent.

b - Area under the curve (AUC)

The (AUC) is not proportional to the administered dose after single IV or oral administration.

The increase in dose results in more than
proportional increase in the AUC due to
saturable drug elimination.

c - The steady state concentration

The steady state drug conc is not proportional to the administered dose during multiple drug administration.

The increase in dosing rate results in more
than proportional increase in the s.s. conc
due to saturable drug elimination.

d - Pharmacokinetic parameters

The estimated pharmacokinetic parameters such as the half life and total body clearance are concentration dependent, and can be different after administration of different doses.
