Clinical significance of pharmacokinetic/pharmacodynamic modeling

Duration of effect :

The pharmacodynamic models can be used to predict the effect of the drug at any time after drug administration. When there is a specific drug conc required to produce the effect, the duration of effect will be the length of time the drug conc remains above this particular conc.

When rapid equilibrium is established between the plasma and the site of action, the pharmacokinetic model alone can be used to predict the duration of effect.

Assume that:
The minimum effective conc for
the drug is 10 mg/L. This means
that the drug will produce its
effect as long as the drug plasma
conc is above 10 mg/L.

Notice that increasing the dose prolongs the duration of effect. However, the duration of effect will not be proportional to the administered dose.
