CLT and Vd (Cont'd)

CLT

Vd
= k =

0.693

t 1/2

When drugs have similar half lives (similar k), this does not mean that these drugs have similar CLT and Vd. This means that the ratios of CLT/Vd for these drugs are similar.

Example:

Ciprofloxacin

CLT = 0.34 L/hr/kg

Vd = 2 L/kg

Chloramphenicol

CLT = 0.17 L/hr/kg

Vd = 1 L/kg

Ciprofloxacin:
k = CLT/Vd = 0.17 hr-1

Chloramphenicol:
k = CLT/Vd = 0.17 hr-1

The plasma conc-time profiles for ciprofloxacin and chloramphenicol decrease at similar rates (similar k, and half life). This is because the ratios of CLT/Vd for both drugs are similar.
