Determination of the Absorption Rate Constant

a- The method of residuals:

This method assumes that drug absorption is first order, ka > k, and the drug follows one-compartment pharmacokinetic model.

When the absorption process is complete, the rate of decline in the linear portion of the curve will be affected by the elimination process only.

e -kat

The residuals are the differences between the y-coordinate values on the extrapolated line and the plasma drug concentrations at the same time points.

The residuals vs. time plot can be expressed as:

Residuals =

F D ka

Vd (ka -k)
e -kat

A plot of the residuals versus time is a straight line that declines at a rate dependent on ka. The slope of this line is -ka/2.303.
